Abstrak

Escherichia coli adalah salah satu bakteri yang dapat mengalami mutasi akibat radiasi sinar ultraviolet. Identifikasi untuk membedakan bakteri yang mengalami mutasi masih cukup sulit dan membutuhkan waktu yang lama. Spektrofotometri FT-IR dapat digunakan untuk identifikasi bakteri secara cepat dan mudah. Penelitian ini dilakukan untuk membedakan bakteri E. coli galur murni dan E. coli galur mutan (akibat radiasi ultravilet) berdasarkan pola spektrum utama dan spektrum derivatif kedua FT-IR. Hasil penelitian menunjukkan pada spektrum utama E. coli galur mutan selalu muncul serapan pada bilangan gelombang 1154 – 1157 cm-1 yang tidak ditemukan pada spektrum E. coli galur murni yang diduga sebagai regang amida II protein. Spektrum derivatif kedua menunjukkan serapan pada bilangan gelombang 2964 –2967 cm-1 yang hanya ditemukan pada spektrum E. coli galur murni, di mana serapan tersebut diduga sebagai regang C-H alifatik.

Abstract

Escherichia coli is one of bacteria which can be mutated by ultraviolet radiation. Identification of mutant bacteria still quite difficult and take a long time. FT-IR spectrophotometry can be used to identificate bacteria with a simple procedure. This research had been done to distinguish the E. coli wild type and E. coli mutant (caused of ultraviolet radiation) by identification of original and second derivative FT-IR spectrum pattern. The result showed that original spectrum of E.coli mutant had a characteristic bands at 1154 – 1157 cm-1, suspected as stretching in amide II protein. Second derivative spectrum gave an absorption at 2964-2967 cm-1 that was only found for E. coli wild type, suspected as the C-H stretching aliphatic.
